Key information: HM Janelia Fluor™ 526, SE is a fluorogenic spontaneously blinking yellow-emitting dye; supplied with an NHS ester reactive group for the labeling of primary amines; hydroxymethyl derivative of JF526. Suitable for live cell imaging. Application: Suitable for confocal microscopy, super-resolution microscopy (SRM) including dSTORM, STED, SIM and single-molecule localization spectroscopy (SmLSM). HM Janelia Fluor™ 526, SE is cell permeable. Properties and Photophysical Data: The spontaneous blinking properties of HM Janelia Fluor™ 526, SE avoids the requirement for strongly reducing dSTORM buffers. NHS ester can be converted to relevant substrate for use in self-labeling tag systems, e.g. HaloTag™ and SNAP-tag™. Excitation and emission maxima (λ) are 526 nm and 550 nm, respectively. To measure the absorbance spectrum of this dye we recommend the following solvent: TFE plus 0.1% TFA.
