Description
- Recognizes human SELP
- SELP (P-Selectin, GMP-140, CD62P) is a transmembrane glycoprotein that mediates interaction of activated endothelial cells or platelets with leukocytes
- Adhesion molecule involved in pathogenesis of inflammation, thrombosis, oncogenesis
- Stored in alpha-granules of platelets and Weibel-Palade bodies of endothelial cells
- Upon cell activation by agonists, P-selectin is transported to cell surface
Process:
- Antibody specific for SELP pre-coated onto microwells
- SELP protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for SELP is added to detect captured SELP protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
